Cell density and solvent are critical parameters affecting formazan evaluation in MTT assay BABT

The aim of this study was to establish the more accurate protocol for fibroblast cell viability using MTT assay. NIH/3T3 fibroblasts were seeded at the following cell densities: 3.125x10³; 1.156x10(4); 3.125x10(4); 1.156x10(5) and 3.125x10(5) cells/cm². Following 24h of seeding, MTT was added to the wells. After 4h of the MTT addition, different solvents were added to solubilize the formazan crystals: 1) HCl/SDS group- 20% SDS and 0.01 M HCl; 2) EtOH/ HAc group-50% ethanol and 1% acetic acid; 3) DMSO group- 99.5% dimethyl sulfoxide; and 4) PropOH group- 99.5% isopropanol. Abstract: The objective of this work was to evaluate different solvents and times in the ultrasound-assisted extraction of polyphenols, in order to quantify the total phenolic and flavonoid contents in and the antioxidant activity of mate (Ilex paraguariensis) leaves. The experiment was carried out in a completely randomized design, in a 8x3 (solvent x time of extraction) factorial arrangement. The solvent extractors were distilled water and aqueous solutions of ethanol, acetone, and methanol, acidified or not with hydrochloric acid, and the time of ultrasound-assisted extractions were 15, 30, and 45 min. Se investigó la influencia de la temperatura (50 y 60 ºC), la relación disolvente-semillas (3:1 y 5:1), la agitación (100 y 300 rev/min) y la concentración de hidróxido de sodio (0,5 y 1,0 N) en la extracción de colorante de bija. La extracción se favoreció con el aumento de la temperatura y la disminución de la concentración de sosa. La agitación prácticamente no causó efectos en los niveles de extracción. Las relaciones disolvente-semillas ensayadas no tuvieron incidencia en la cantidad de pigmentos extraída.

Highly Solvent Tolerance in Serratia marcescens IBBPo15 BABT

ABSTRACT The aim of this study was to investigate the solvent tolerance mechanisms in Serratia marcescens strain IBBPo15 (KT315653). Serratia marcescens IBBPo15 exhibited remarkable solvent-tolerance, being able to survive in the presence of high concentrations (above 40%) of toxic organic solvents, such as cyclohexane, n-hexane, n-decane, toluene, styrene, and ethylbenzene. S. marcescens IBBPo15 produced extracellular protease and the enzyme production decreased in cells exposed to 5% cyclohexane, n-hexane, toluene, styrene, and ethylbenzene, as compared with the control and n-decane exposed cells. Implementation of Policy Instruments for Chlorinated Solvents: A Comparison of Design Standards, Bans, and Taxes to Phase Out Trichloroethylene AgEcon

This paper studies the Swedish prohibition of the hazardous solvent Trichloroethylene (TCE). Sweden is alone in completely prohibiting its use. The ban has been at best a partial success and illustrates the dilemmas of policymaking. Use has declined but not stopped, largely because the decision to ban TCE was challenged in the courts. Recently, the EU Court of Justice decided in favor of Sweden's right to have a ban. This article analyzes abatement cost data to show that the cost of replacing TCE is low for most plants, although there appear to be a few firms for which it may be quite high. Interference of propylene glycol with the hole-board test BJMBR

Experimental drugs and/or plant extracts are often dissolved in solvents, including propylene glycol. Nevertheless, there is evidence for psychoactive properties of this alcohol. In this study we found that in the hole-board test 10% propylene glycol did not modify the head-dipping behavior. However, 30% propylene glycol induced an increase in the number of head-dips (46.92 ± 2.37 compared to 33.83 ± 4.39, P<0.05, ANOVA/Student-Newman-Keuls), an effect comparable to that obtained with 0.5 mg/kg diazepam (from 33.83 ± 4.39 to 54 ± 3.8, P<0.01, ANOVA/Student-Newman-Keuls). These results demonstrate that 30% propylene glycol has significant anxiolytic effects in this model and therefore cannot be used as an innocuous solvent.

Response of Rhodococcus erythropolis strain IBBPo1 to toxic organic solvents BJM

Abstract Recently, there has been a lot of interest in the utilization of rhodococci in the bioremediation of petroleum contaminated environments. This study investigates the response of Rhodococcus erythropolis IBBPo1 cells to 1% organic solvents (alkanes, aromatics). A combination of microbiology, biochemical, and molecular approaches were used to examine cell adaptation mechanisms likely to be pursued by this strain after 1% organic solvent exposure. R. erythropolis IBBPo1 was found to utilize 1% alkanes (cyclohexane, n-hexane, n-decane) and aromatics (toluene, styrene, ethylbenzene) as the sole carbon source. The influence of organic solvents on estimates of genotoxicity and antigenotoxicity in the SOS chromotest Genet. Mol. Biol.

In this work, the toxicity and genotoxicity of organic solvents (acetone, carbon tetrachloride, dichloromethane, dimethylsulfoxide, ethanol, ether and methanol) were studied using the SOS chromotest. The influence of these solvents on the direct genotoxicity induced by the mutagens mitomycin C (MMC) and 4-nitroquinoline-1-oxide (4-NQO) were also investigated. None of the solvents were genotoxic in Escherichia coli PQ37. However, based on the inhibition of protein synthesis assessed by constitutive alkaline phosphatase activity, some solvents (carbon tetrachloride, dimethylsulfoxide, ethanol and ether) were toxic and incompatible with the SOS chromotest.
